Assistant manager Tommy Wright and goalkeeping coach Mark Crossley have been placed in temporary charge, BBC report.
The move follows a disappointing start to the new campaign for Chesterfield who were relegated from League One last season. The Spireites are winless from four competitive games so far and currently lie 18th.
Following three seasons at his old club Oldham Athletic, Sheridan moved south in June 2009 and led Chesterfield to the League Two title and promotion in his second season in charge.
Their stay in League One was fleeting though and they were relegated in 22nd place at the end of last season.
During his playing days, Stretford-born Sheridan won 34 caps for Ireland including appearances at the 1990 and 1994 World Cups.
